(NH4) 2SO4 (36%) 5.0 ml

MgSO4 0.5 g

Distilled Water up to 1.0 L

Adjust pH to 6.8-7.0

Trace elements:

EDTA 500.0 mg

FeSO4.7H2O 200.0 mg

ZnSO4.7H2O 10.0 mg

MnCl2.4H2O 3.0 mg

H3BO3 30.0 mg

CoCl2.6H2O 20.0 mg

CuCl2.2H2O 1.0 mg

NiCl2.6H2O 2.0 mg

Na2MoO4.2H2O 3.0 mg

Distilled Water 1.0 L

Dissolve ingredients in the order given.

Medium made up as follows:

Yeast extract 1.0 g

Bacto Peptone 5.0 g

Lactose 10.0 g

Agar 15.0 g

Basal salts 1.0 L

Autoclave at 121 °C for 15 minutes. Then add aseptically,Lactose and 1ml trace elements after sterilization.
