Beef liver (cut up in small pieces) 500.0 g

Tap water 1.0 L

Soak beef liver overnight in refrigerator. Skim fat off top. Autoclave for 10 minutes at 15 lbs. Filter through cheesecloth save meat. Add:

Peptone 10.0 g

K2HPO4 1.0 g

Adjust pH to 8.0. Filter through paper and make up volume to one liter with more tap water. Place 5 mg CaCO3 in each tube; add meat to a depth of 1/2 inch; cover with broth to a total depth of 2 inches.
